Bone marrow samples from AML patients possess increased GST-FLT3S phosphorylation activities. A. White blood cells from two normal and four AML bone marrow samples were extracted. Kinase assays were performed with cell extracts at 0.2 mg/ml in the presence of 0.2 mg/ml GST-FLT3S. Phosphorylation of GST-FLT3S was detected by using anti-phosphotyrosine, and equal protein loadings were revealed by anti-GAPDH (for house-keeping protein glyceraldehyde 3-phosphate dehydrogenase) and Coomassie blue staining (for GST-FLT3S).
