Intracellular uptake and retention assay. a Intracellular incorporation of C-14 bosutinib is reported as the number of hits obtained from beta-counter. The results derive from three independent experiments ± SD. For each drug transporter, the statistical difference between overexpressing or silenced cells and K562S was calculated with two-tailed unpaired student’s t-test, and a p value of 0.05 was chosen as the limit of statistical significance (** = p < 0.05 *** = p < 0.01 NS, not significant). b C-14 bosutinib intracellular accumulation after drug transporters inhibition. For each transporter, the cells were pre-treated with specific drug-transporter inhibitors. C-14 bosutinib incorporation obtained with the pre-treatment was normalized over C-14 bosutinib incorporation in the same cell line without the pre-treatment. Results are an average of three independent experiments.
