Effects of GMR CAR T cells generated from two patients with JMML on the colony growth of JMML CD34+ cells isolated at the time diagnosis. a Effect of mock T cells or GMR CAR T cells generated from pre-transplant PBMCs in a patient with an NRAS mutation who received no chemotherapy (patient no. 3) on the colony growth of corresponding CD34+ cells isolated at disease onset were evaluated. The inhibitory effects of GMR CAR T cells were compared between the patient and a healthy donor. b Effects of mock T cells or GMR CAR T cells generated from a patient with both PTPN11 and SETBP1 mutations who achieved complete chimerism after cord blood transplantation from an unrelated donor (patient no. 4) on the colony growth of corresponding CD34+ cells isolated at disease onset were evaluated. The inhibitory effects of GMR CAR T cells were compared between the patient and a healthy donor. Values are expressed as percentages of the total colony numbers obtained by culture in the absence of T cells. The total numbers of colonies grown by JMML PB CD34+ cells in the absence of T cells were 71.3 ± 2.3 in patient no. 3 and 55.3 ± 5.1 in patient no. 4
