Effects of GMR CAR T cells derived from a patient with JMML and healthy donors on the proliferation of JMML CD34+ cell populations. PB CD34+ cells (1 × 104) isolated from a patient with PTPN11 mutation and trisomy 8 (patient no. 6) were cultured on irradiated AGM-S3 cells with or without 1× 104 mock T cells or 1 × 104 GMR CAR T cells in 10 % FBS-containing culture supplemented with 10 ng/ml SCF plus 10 ng/ml TPO. After 7 days, cultured cells were stained with APC-conjugated anti-CD34 and PE-conjugated anti-CD38 mAbs. The addition of both JMML patient-derived GMR CAR T cells and healthy donor-derived GMR CAR T cells reduced the number of JMML CD34+CD38 cells compared to culture in the absence of T cells or culture in the presence of mock T cells
