TSP-2-induced MMP-2 activation and migration were mediated through CD36 and integrin αvβ3 in human PCa cells. Starved PC-3 (open bar) and DU145 (closed bar) cells were pretreated with SSO (500 and 5000 nM), RAD control peptide (200 nM), RGD peptide (200 nM), and BIO1211 (5 and 500 nM) for 1 h, followed by incubation of TSP-2 for another 24 h. The abilities of migration (a), invasion (b), as well as the expressions of MMP-2 protein (c), mRNA (d), and its activity (e) were assessed by the transwell assay, immunoblotting, real-time PCR, and zymography, respectively. Cells without treatments were used as a control (set to 100), and data were shown as multiples of that. Results are shown as the mean ± SEM (n ≥ 3, *p < 0.05 when compared to untreated control; # p < 0.05 when compared to the TSP-2-treated group)
