A novel working model for caspase-1-mediated gene expression. (I) In response to DAMPs and PAMPs, caspase-1 activates IL-1β and IL-18 which in turn will magnify the inflammation by enhancing secondary pro-inflammatory genes. (II) Caspase-1 may get localized to the nucleus in response to stress leading to degradation of histone modifier Sirt-1. This may lead to activation of genes that are suppressed by Sirt-1. (III) Caspase-1 can regulate gene expression by mechanisms that are independent to the mechanisms mediated by its downstream targets IL-1β, IL-18, and Sirt-1
