From: Pharmacological targeting of the ephrin receptor kinase signalling by GLPG1790 in vitro and in vivo reverts oncophenotype, induces myogenic differentiation and radiosensitizes embryonal rhabdomyosarcoma cells

Fig. 6

GLPG1790 radiosensitizes ERMS cell lines and modulates signal transduction pathways. RD and TE671 cells untreated or pretreated with GLPG1790 for 24 h were exposed or not to 4 Gy radiation treatment (RT). Two hours after RT, cells were seeded at low concentration for colony assays or lysed for total protein extraction. a Representative pictures of colonies stained with crystal violet after 14 days of GLPG(−)/RT(−), GLPG(−)/RT(+) or GLPG(+)/RT(+) treatments (Upper Panel). Colony forming efficiency was calculated by crystal violet absorbance (Lower Panel). Results represent the mean values ± SD of three independent experiments. Statistical significance: ***p < 0.001 vs. GLPG(−)/RT(−), arbitrarily set at 1; $$$ p < 0.001 vs. GLPG(−)/RT(+). b Cell lysates from RD and TE671 cells were analysed by immunoblotting with specific antibodies for indicated proteins; α-Tubulin expression shows the loading of samples. Representative of three independent experiments. c Cell lysates from RD and TE671 treated ± 3.5 μM GLPG1790 for different times were analysed by immunoblotting with specific antibodies for indicated proteins; α-Tubulin expression shows the loading of samples. Representative of three independent experiments
