    It has recently been reported that humanized AML constitutive activation of canonical Wnt signal pathway not in the hematopoietic stem cells (HSCs) but in osteoblastic niche cells (doi:10.1038/bonekey.2014.91). Accumulation of beta-catenin in the nucleus in the niche cells induce the secretion of Notch ligand Jagged-1, thereby blasts with chromosomal instability derived from HSCs selectively proliferate with enhanced the signal crosstalk between canonical Wnt and Notch signal. Although, the precise mechanism of abnormal osteoblastic niche causes the chromosomal translocation which leads to the development of MDS/AML, the aberrant regulation of canonical Wnt signal pathway might be a good therapeutic target for MDS/AML.
